I currently have the SugarCRM 5 1.1.18 jumpbox running on ESXi 4 server. Can I upgrade to SugarCRM 6.0 using the upgrade files from the SugarCRM site, or do I have to wait until JumpBox develops a new appliance for that version?

As always, you're welcome to try anything you want with a JumpBox. And as always, I *strongly* suggest you not run any experiments on your production JumpBox. If you do use Sugar's upgrade, things in the JumpBox may break, in particular the backup/restore.
As to SugarCRM 6, I expect a JumpBox for SugarCRM6 on Ubuntu 10.04 (Lucid Lynx) to be released in a couple weeks. Not sure at the current time if it will be released as a beta (because of both the new version of Sugar as well as lucid).
